Lineage, Twin & Avuncular DNA Tests
Explore deeper family connections — twin zygosity, aunt/uncle relationships, and paternal or maternal lineage — privately, for your own peace of mind.
Not every family question fits a standard paternity test. This collection covers the relationships that need a different approach: confirming whether twins are identical or fraternal, estimating an aunt/uncle (avuncular) relationship when a parent isn't available, and tracing paternal (Y-chromosome) or maternal (mtDNA) lineage across generations.
Lineage tests work differently from relationship tests. The Y chromosome and mitochondrial DNA are passed down nearly unchanged along a single line, so they confirm a shared line rather than an exact one-to-one relationship. As with everything in this section, these tests are for personal peace of mind, not legal use.
Lineage & Twins tests

Twin Zygosity Test
Twins (or their parents) curious whether they're identical or fraternal
- Participants:
- Two Twins
- Lab time:
- 2 business days

Avuncular Test (Aunt / Uncle)
An aunt or uncle and a child estimating a biological relationship
- Participants:
- Aunt or Uncle, Child (mother optional)
- Lab time:
- 5 business days

Y-Chromosome (Male Lineage) Test
Two or more males checking whether they share a paternal line
- Participants:
- Two or more males
- Lab time:
- 5 business days

Mitochondrial DNA (Female Lineage) Test
Two or more people checking whether they share a maternal line
- Participants:
- Two or more people
- Lab time:
- 14 business days
How it works
- 1
Order your kit
Choose your test and check out online. No appointment or doctor's order needed.
- 2
We ship within 1 business day
Your collection kit ships free with cheek swabs, step-by-step instructions, and a return mailer.
- 3
Collect samples at home
Each participant gently swabs the inside of their cheek. Painless and takes about a minute per person.
- 4
Mail samples to the lab
Drop your samples in the mail using the supplied envelope. Prepaid Return Mailer or FedEx Overnight Label Provided. Lab processing time starts when the lab receives your samples.
- 5
Receive results securely
The lab sends your report to us and we forward it to you privately by email.
Who uses this test
This group serves a range of personal questions:
- Parents of twins who want to settle whether their twins are identical or fraternal.
- An aunt or uncle and a child estimating a biological link when an alleged parent isn't available.
- People researching family history who want to confirm a shared paternal or maternal line — for example, two men checking a common surname line, or relatives confirming a maternal connection.
Each test can answer its specific question for your own information. None of them can be used for legal purposes, and the lineage tests in particular confirm a line, not a precise relationship — they can't, on their own, tell you exactly how two people on the same line are related.
What you’ll need
- The cheek swabs included in your kit — no needles, no blood
- The right participants for your question (e.g. both twins; an aunt/uncle and child; males on a paternal line for Y-DNA)
- Patience for mtDNA testing, which takes longer to process
- Consent from every adult participant
Accuracy & methodology
Each test uses the method suited to its question:
- Twin zygosity compares many markers — identical twins match across all of them; fraternal twins differ like ordinary siblings. This is highly conclusive.
- Avuncular testing reports a relationship likelihood (a combined index and probability); including the child's known parent strengthens it.
- Y-chromosome testing compares the male-line signature shared by fathers and sons; a match indicates a shared paternal line.
- mtDNA testing compares the maternal-line signature everyone inherits from their mother; a match indicates a shared maternal line.
Because Y-DNA and mtDNA are shared by everyone on the line, they confirm a shared lineage rather than a specific relationship. Testing is performed by our accredited partner laboratory.
Testing is performed by our AABB-accredited partner laboratory.
Frequently asked questions
- Can a lineage test tell me my exact relationship to someone?
- No. Y-chromosome and mtDNA tests confirm whether you share a paternal or maternal line. Because everyone on that line shares the same signature, they can't, on their own, distinguish (for example) a brother from a cousin on the same line.
- Who can take a Y-chromosome test?
- Only males carry a Y chromosome, so the test compares two or more males on the paternal line of interest. Women would need a male relative on that line to participate.
- Can men take an mtDNA test?
- Yes. Everyone inherits mtDNA from their mother, so both men and women can be compared on a maternal line — though only women pass it on.
- How reliable is a twin zygosity test?
- Very. Identical twins match at every marker tested, while fraternal twins show ordinary sibling-level differences, so the result is highly conclusive.
- Why does mtDNA testing take so long?
- mtDNA analysis sequences a different part of the genome and is more involved, so lab processing takes around 14 business days from when the lab receives your samples — longer than our other tests.
- Are any of these admissible in court?
- No. Every test in this section is for personal peace of mind only and cannot be used for legal matters.
Related tests & legal note
If you're trying to confirm a close, specific relationship rather than a line, a paternity, siblingship, or grandparentage test will serve you better. Curious about deep ancestral origins instead? See our GPS Origins ancestry test under Lifestyle & Wellness.
Need a legal result? None of these tests can be used in court or for immigration. Those require a court-approved, chain-of-custody DNA test through an accredited laboratory.
Need results for a custody case, child support, immigration, or any other legal matter? An at-home test cannot be used for those purposes. You'll need a court-approved, chain-of-custody DNA test performed through an accredited laboratory, where a neutral third party verifies every participant's identity and handles the samples.
